建於1860年,由土生葡人馬奇士設計,原稱為"伯多祿五世劇院"(伯多祿五世為葡萄牙國王,於1853年至1861年在位),
1860年只建成主體部分,至1873年才加建具新古典主義建築特色的正立面,設置276個座位.
崗頂劇院是中國第一所西式劇院,亦是當年葡人社群舉行重要活動的場所.
